Hello everyone,

has anyone already used GCaMP8 with the miniscopes?
We are especially interested in the m version. If so, could you share some details regarding your experience, titer, region etc?

Hi Florian,

If I'm not mistaken, reportedly the GCaMP8 virus is better then previous virus versions.

As for my experience, I use also GCaMP8 of more GCaMP virus and I'm satisfied with it.
The cells activity just is good than other types. I didn't notice any better or worse difference in the imaging and histology.
My titer is 1:5, but someone use from 1:4 to 1:8 ratio. I think it doesn't matter especially.

In the aspect of my research topic, I have a Miniscope V4.4 and the GRIN lens properties are 4mm length and 1mm diameter.
I investigate the dentate gyrus (DG) and CA1 in the hippocampus.

Hi Eric,

thanks for your experience, helps a lot to hear! Could you tell me what original titer your virus has before you dilute it 1:5?

We are currently using GCaMP7f in our lab (and record in mPFC) and would be very interested to see if it is worth it to "upgrade" the GCaMP. Our main concern is that GCaMP8m might have quite fast dynamics for miniscopes. How many frames per second do you record usually?

Would it be possible to look at a short snippet of your recordings so that we can compare it lab internally to our GCaMP7f videos?

I’ve tried both 7f and 8m in PFC, and ultimately went back to 7f after trying different 8m dilutions. 8m had such strong signal that, in my hands, the higher background fluorescence diminished overall quality. I’m sure I could have done better to optimize the 8m surgery but 7f was sufficient
