Question on Starting (First Project?)

Hi! This is my first post, so I hope I don't come across as stupid around here (I know there are quite a few professional people here, it's quite intimidating.) I'm currently in highschool and have learned a lot about dna extraction and the process of making recombinant dna for transgenic organisms, (vectors, restriction enzymes, etc), but I have yet to learn the inside of how you'd do it in a diy-lab setting. I was only interested in doing a basic project with making recombinant dna (bio-luminescent bacteria, etc.) Could anyone help me out, or at least put me in the right direction. This has greatly interested me in the past week, and I'd like to look into it :)

you do not come across as stupid, so don't worry about that.

Question: have you actually done all of this in class, or have you just studied it?  If you have done it in class...well thats how we do it as well (mostly).

If you have not, check out www.bosslab.org to see protocols we use at BOSSLAB, and if you want to see some cool pictures you can check out www.tequals0.wordpress.com.  If you are in the boston area, come down to a meeting!  we have a meeting saturday from 12-1 pm at BOSSLAB (just look it up on google maps).  Let me know if you need help getting a hold of equipment or supplies.

I'm near Baltimore, so traveling to Boston from here would probably be a bit of a stretch. The only thing we did in class was separating dna from pureed bananas. It was an interesting lab, and we went into the basics of how transgenic organisms are made, but it wasn't very specialized, and we didn't have any labs on that specifically, which is why I took interest in this, to take what I learned one step further. And thanks for the link, I'll check it out :)

Oh.

Well making glowing/green bacteria is not hard.  I will put instructions up sometime tonight, I was typing them up since I am chilling in the lab tonight, working on yet another protocol.

Okay, awesome! I saw the Bosslab post on their gfp-E.coli, really neat. I figured it was one of the easier things that could be done considering it's only bacteria, and how many people have done it .

Just read the protocol post. It doesn't seem too complicated, the main adventure for me really will be finding out how get my hands on supplies (quite a few of which I think I can find at my high school or the STEM high school my friends go to.)
