What is the cheapest and cleanest way to make a work hood that at
least two people could look over your shoulder at what you're doing
while working, 40 to 60cm deep and that much if not more wide, at
least clean and strong enough to leave cooling agar plates open inside
while on?

I know the big panel filters are at least $100-150 USD, and a copper
or stainless grating is probably a good (but not cheap) idea for a
vent to protect the filter or cover the air duct.

For a while I just used a big HEPA air purifier attached to a clear plastic crate through terribly-sawn openings. It was very positive pressure: light items would be blown out! But, it worked well until one day it didn't; I believe the filter was finally saturated.

In my experience, then, a good pos-pressure hood is trivial and mobile, but I use a bunsen exclusively these days.

Depends on what you have.  If you can weld etc. Then thats one way, but I recently saw some real laminar flow hoods on sale for 500 used.

So the burner really cleans the air? Seems like the convection current
would just swirl dirty air around more...

Use a cheap houes type air filter as a pre-filter to extend the life of the expensive HEPA.
