Usually autocomplete fires up automatically when a . or ( is typed. Looking at the code I can't immediately say how a completion is implemented (other than "it looks like it is provided by rope"), so I guess somebody should describe that and/or implement debug hooks that print what's going on with completion when Ctrl-Space is clicked and doesn't work.

To debug introspection features like autocomplete, go-to-definition or
calltips, one has to set the SPYDER_DEBUG environment variable to 1.

So, in a bash terminal, type the following before re-executing Spyder:
$ export SPYDER_DEBUG=1

Then, two more files should appear in your ~/.spyder2 directory:
'monitor_debug.log' and 'rope.log'.
They contain very useful informations to solve this problem.
