Some of you at the meeting saturday heard me talk about the bluegene project, basically we want to take a gene from one bacteria and transfer it to another bacteria.  This gene codes for an enzyme that breaks down indole, and turns it into indigo.  Right now, the team is Cat, Simon, and myself.If you want to join, please do so this week, by Friday.  The cost of the project is basically whatever we buy, divided between all the people in the project.  I estimate that if nothing goes wrong, cost will be under $200 per person.

Hey,

The bacteria produces the pigment from scratch (glucose)? No need for external substrate??

Well, in that case, glucose would be the substrate, wouldn't it? :p

The indigo is produced by breaking down indole, which e. Coli naturally makes.

Surely, you're right. but e.g. the plasmid that makes e.coli smell like vanilla needs ferulic acid as a substrate. Who the hell has ferulic acid in his kitchen? ;D

As far as I know, you can just grow them on LB, but yes sometimes you need to add chemicals to get weird smells/colors.

As someone who is interested in molecular biology and organic chemistry (and with a bit of background in dye chemistry), I think this is a really cool project. Unfortunately, I am nowhere near you guys, nor do I have the money to put into this. I am excited to see how this goes. Keep us updated!

So I assume, all bacteria and animals have indole? Plants?


So you could also make a blue-skinned dog by attaching an animal promotor etc.  :D

or anabolizing tryptophan

Except the promoter must not be too strong or the animal could become
deficient in tryptophan... also maybe the dye has some toxicity/effect
in multicellular organisms
