reconstructing non-CTF corrected tomograms

Hi everyone!


I’d like to reconstruct my tomograms with WarpTools, but do the CTF correction using a different, external software.

What’s the best way to do this? Can I just skip ts_ctf and run ts_reconstruct after tilt alignment or is there a specific flag I can use in e.g. ts_reconstruct?

I assume you’re looking into exporting your data to cryoDRGN? If yes please let me know how it goes )) to answer your question: yes, you can. I did it accidentally, while missing the ts_ctf step.

Hi Karin,

thanks for the replay! :) It's actually for denoising with IsoNet2, but I would like to give cryoDRGN a go at some point as well- will let you know how it goes when I try it.

Hi,

I dont know what happens if you miss the ts_ctf step. But I used WarpTools half-maps for isonet and got pretty good results. 

I think in isonet2 you can specify that the tomogram is ctf corrected anyways

thanks for replying! Yes, you're completely right, you can inactivate isonet2's own ctf correction. I just wanted to see how ctf correction in isonet2 compares to warp. :)
