With the Pod HD500 is it possible to turn off one effect block while
turning on another simultaneously?

Sure, you only need to assign a pedal to the effect block and switch
it off by default in the patch. When you then kick the pedal, you
switch off on effect (or a series) and switch on others. I use it all
the time to play U2 patches since you cannot switch from one patch to
another without loosing the delay trails. (so you need one patch with
all effects in their 'starting' position). What you cannot do is
assign 2 pedals to 1 effect. It would allow me to switch off a
specific effect during 1 transition and switch it back on in another
transition where other effects are added too.

forgot to add that may need to go in the setup menu and change your
footswitch mode from abcd to something else (FS5-8 in my case but I
have a bean, not the HD500). abcd is the setting to control every
effect individually.

maybe you can take a look at Stilwel's stock plugin baby preset to see
this. I think quite a few FX are turned off/on when you use the FS1 or
FS2 switch
