Estimation of BCR clonality

Hi Bo and Xiahao,

your paper on the landscape of B-cell immunity recently was really interesting. Fantastic job! 

I was wondering what your thoughts on estimation of BCR clonality using the output that TRUST provides are? You mentioned in an earlier post that the frequencies are estimates that don't necessarily track with the actual numbers due to sampling noise. Do you have any guidance on what number a decent coverage constitutes? And do you think there is value in trying to estimate BCR clonality using the TRUST output?
