Blade teams up with the Nightstalkers, led by Abigail Whistler, the daughter of his mentor and friend Abraham Whistler, who was killed by the FBI agents who raided Blade's hideout. The Nightstalkers have developed a bioweapon called Daystar, which can kill vampires at the genetic level, but they need a pure blood source to make it effective. Blade and the Nightstalkers must find Drake before he can be used by Danica and her allies to create a new breed of vampires that can walk in daylight.

The film was written and directed by David S. Goyer, who also wrote the screenplays for the first two Blade films. Goyer had originally planned to make a prequel film about Blade's origins, but decided to make a sequel instead after Snipes expressed interest in returning to the role. Goyer wanted to introduce new characters and villains to expand the Blade universe and appeal to a wider audience. He also wanted to make the film more comedic and lighter in tone than the previous films. He cast Reynolds and Biel as Hannibal King and Abigail Whistler, two members of the Nightstalkers who provide comic relief and romance respectively. The DVD also included several bonus features, such as audio commentaries by Goyer and the cast, deleted scenes, alternate endings, blooper reel, featurettes on the making of the film, visual effects, stunts, weapons and costumes, galleries of concept art and storyboards, music videos and trailers. The Blu-ray version included all of these features plus an interactive trivia game.
