Yes, TIQAM discussion site is still active.

Regarding software choice for MRM, there are vendor specific commercially
available software. TIQAM is a good open source software for generating
"theoretical" and/or observed (in PeptideAtlas) as well as visual inspection
of your transitions once you measured the transitions.

You can download TIQAM software/tutorial and dataset from
