The first season opens with Peter Parker entering his junior year at Midtown Manhattan Magnet High School, having only been bitten by a genetically-enhanced spider and acquired his powers in recent months. Peter struggles to balance his responsibilities as the costumed hero Spider-Man with the problems of his personal life. In addition to fighting crime, he must navigate his romantic affections for Gwen Stacy and Mary Jane Watson; maintain his friendships with Harry Osborn and Eddie Brock; deal with being bullied by football star Flash Thompson at school; keep his internship position as a lab assistant to Dr. Curt Connors at Empire State University; and help to support his Aunt May after the death of his Uncle Ben by working as a freelance photographer at the Daily Bugle. The Bugle is owned by loudmouth publisher J. Jonah Jameson, who often uses Peter's pictures of his alter-ego to discredit and carry out a smear campaign against Spider-Man.

Created by Stan Lee and Steve Ditko, Spider-Man first appeared in the anthology comic book, Amazing Fantasies #15 in 1962 and has since been featured in films, TV shows, novels, video games, and plays. For those not familiar, "Spider-Man" is the alias of Peter Parker, an orphan raised by his Aunt May and Uncle Ben in New York City after his parents died. In his origin story, Parker gets superhuman spider powers and abilities from a bite from a radioactive spider; these include clinging to surfaces and ceilings, superhuman strength, and speed. He can also detect danger with his precognition ability called "spider-sense." After the personal tragedy of his Uncle Ben, Peter began using his spider powers to fight against crime.
Returning to ESU, Peter is confronted by Debra's boyfriend Biff Rifkin for how he blew up at Debra the day before. Peter simply knocks him out with his spider-strength before being pulled into Professor Sloan's office. Sloan suggests that Peter gives up his Teachers Assistant job so that he can focus on his studies, a decision Peter concedes to. He abruptly leaves, unaware that Debra is following him. When he suddenly disappears and Spider-Man is seen swinging by, Debra begins to suspect that Peter is Spider-Man.
Black Cat is one of the show's best villains, and for good reason. She's easily able to keep up with Spider-Man's quips and barely sees him as a threat, stating "Cats don't fight spiders. We just bat them around." She proves this correct, as she later steals expensive jewelry right under Spider-Man's nose.
Peter Parker AKA Spider-Man is the primary protagonist of The Spectacular Spider-Man. During a field trip in high school student, he was bitten by a radioactive spider. Seeing as he had attained immense power, Peter entered a cage match against Crusher Hogan, in which he was promised $1,000 should he win. Unsurprisingly, Parker won the match, but was denied the full prize money due to his refusal to sign onto a management contract. Frustrated at his wasted efforts, he purposefully let a criminal by the name of Walter Hardy pass by when the latter stole the cash kept at the wrestling stadium. Peter would come to realize the magnitude of his actions when his Uncle Ben was murdered following this event. Upon tracking down the killer, it was revealed to be Walter Hardy, the man he could have easily stopped. Realizing his terrible error, Peter went on to fight crime under the alias of Spider-Man, operating by Ben's principle of "With great power, there must also come great responsibility".
