A concierge of an apartment in Barcelona named César is on the roof of a building explaining his nature of not being able to find any happiness no matter what he does or what good things happen to him. As he wakes up in Clara's apartment and begins his routine working in the main lobby, he gives hush money to teen Úrsula, the daughter of another tenant. César visits his sick mother in the hospital, who is practically unable to speak to her son but listens to him talk about Clara. Verónica, another tenant, asks César to take care of her dogs giving detailed directions on how to feed them. César ignores her instructions and feeds her dog Rocío a slice of pie. Clara, entering her apartment, opens up letters she has received then tears them up and throws them away all. While this is happening, César hides under her bed waiting for her to fall asleep in order to use chloroform to keep her asleep so he can maliciously mess with her care product. The morning after, he sneaks out of Clara's apartment with Úrsula waiting outside. She demands that César provide her with an adult movie if he wants her to keep her mouth shut. César, working in the lobby, finds Verónica heading out of the apartment. He questions Verónica where Rocío is and she explains that Rocío has horrible diarrhea from something he may have eaten.
César, once again hiding under Clara's bed, uses chloroform on her to keep her in a deep sleep and begins messing with more of Clara's items and plants bug eggs in her apartment. A couple of days pass and Clara's rashes which she was developing due to César messing with her care products have slightly gone away, relieved for a second she finds many cockroaches in her apartment and decides to stay with her mother until César fumigates her apartment. The police get involved to figure out who is harassing Clara and César frames the cleaner's son which leads to his arrest.

Despite an elevated recovery need, research indicates that athletes often exhibit relatively poor sleep. Timing and consolidation of sleep is driven by the circadian system, which requires periodic light-dark exposure for stable entrainment to the 24-hour day, but is often disturbed due to underexposure to light in the morning (e.g. low-level indoor lighting) and overexposure to light in the evening (e.g. environmental and screen-light). This study examined whether combining fixed sleep schedules with light regulation leads to more consolidated sleep. Morning light exposure was increased using light-emitting goggles, whereas evening light exposure was reduced using amber-lens glasses. Using a within-subject crossover design, twenty-six athletes (14 female, 12 male) were randomly assigned to start the intervention with the light-regulation-week or the no light-regulation-week. Sleep was monitored by means of sleep diaries and actigraphy. Due to low protocol adherence regarding the fixed sleep-wake schedules, two datasets were constructed; one including athletes who kept a strict sleep-wake schedule (N = 8), and one that also included athletes with a more lenient sleep-wake schedule (N = 25). In case of a lenient sleep-wake schedule, light regulation improved self-reported sleep onset latency (Δ SOL = 8 min). This effect was stronger (Δ SOL = 17 min) and complemented by enhanced subjective sleep quality in case of a strict sleep-wake schedule. None of the actigraphy-based estimates differed significantly between conditions. To conclude, light regulation may be considered a potentially effective strategy to improve subjective sleep, but less obtrusive methods should be explored to increase protocol compliance.

In a new study, published today in the journal Nature Communications, researchers at Bar-Ilan University in Israel reveal a novel and unexpected function of sleep that they believe could explain how sleep and sleep disturbances affect brain performance, aging and various brain disorders.
Using 3D time-lapse imaging techniques in live zebrafish, the researchers were able to define sleep in a single chromosome resolution and show, for the first time, that single neurons require sleep in order to perform nuclear maintenance.
The role of sleep is to increase chromosome dynamics, and normalize the levels of DNA damage in each single neuron. Apparently, this DNA maintenance process is not efficient enough during the online wakefulness period and requires an offline sleep period with reduced input to the brain in order to occur. "It's like potholes in the road," says Prof. Lior Appelbaum, of Bar-Ilan University's Mina and Everard Goodman Faculty of Life Sciences and Gonda (Goldschmied) Multidisciplinary Brain Research Center, who led the study. "Roads accumulate wear and tear, especially during daytime rush hours, and it is most convenient and efficient to fix them at night, when there is light traffic."
Appelbaum calls the accumulation of DNA damage the "price of wakefulness." He and his doctoral student David Zada, first author of the study, as well as co-authors, Dr. Tali Lerer-Goldshtein, Dr. Irina Bronshtein, and Prof. Yuval Garini, hypothesized that sleep consolidates and synchronizes nuclear maintenance within individual neurons, and set out to confirm this theory.
Their discovery was achieved thanks to the characteristics of the zebrafish model. With their absolute transparency, and a brain very similar to humans, zebrafish are a perfect organism in which to study single cell within a live animal under physiological conditions. Using a high resolution microscope, the movement of DNA and nuclear proteins within the cell -- inside the fish -- can be observed while the fish are awake and asleep. The researchers were particularly surprised to find that chromosomes are more active at night, when the body rests, but this increased activity enables the efficiency of the repair to DNA damage.
The results establish chromosome dynamics as a potential marker for defining single sleeping cells and propose that the restorative function of sleep is nuclear maintenance. "We've found a causal link between sleep, chromosome dynamics, neuronal activity, and DNA damage and repair with direct physiological relevance to the entire organism," says Prof. Appelbaum. "Sleep gives an opportunity to reduce DNA damage accumulated in the brain during wakefulness."
"Despite the risk of reduced awareness to the environment, animals -- ranging from jellyfish to zebrafish to humans -- have to sleep to allow their neurons to perform efficient DNA maintenance, and this is possibly the reason why sleep has evolved and is so conserved in the animal kingdom," concludes Prof. Appelbaum.
