The specificity does not change. However, if you want to disrupt your gene of choice, it would be best to target the TALEN to the start of a gene (i.e. exon 1). That way when a mutation happens the higher the possibility that the indels will throw you protein out frame early and cause a knock out rather than a dominant negative protein.
