We recently found that nucleosomes directly block access of CRISPR/Cas9 to DNA (Horlbeck et al., 2016). Here, we build on this observation with a comprehensive algorithm that incorporates chromatin, position, and sequence features to accurately predict highly effective single guide RNAs (sgRNAs) for targeting nuclease-dead Cas9-mediated transcriptional repression (CRISPRi) and activation (CRISPRa). We use this algorithm to design next-generation genome-scale CRISPRi and CRISPRa libraries targeting human and mouse genomes. A CRISPRi screen for essential genes in K562 cells demonstrates that the large majority of sgRNAs are highly active. We also find CRISPRi does not exhibit any detectable non-specific toxicity recently observed with CRISPR nuclease approaches. Precision-recall analysis shows that we detect over 90% of essential genes with minimal false positives using a compact 5 sgRNA/gene library. Our results establish CRISPRi and CRISPRa as premier tools for loss- or gain-of-function studies and provide a general strategy for identifying Cas9 target sites.
I want to know which section is currently active. I tried the usePathname as suggested in the docuemntation but this only shows the activa page not the section within the page:
Use useRouter from next/router in Next.js for internal links with hash fragments to access the full path, including the hash, as usePathname from next/navigation only provides the current pathname without the hash.
I was making a simple navbar in a side project and faced a problem with active links in Next.JS, in react you could just use react-router-dom that has a Link component with a activeclass atribute, but not in Next.JS (I think)
I basically did this answer because I was searching about this for 30min or so and didn't find anything simple and actually fast enough for a navbar, if there was a similar or simpler answer please link it to me.
Hi everyone, I generate a report quite frequently for all active project for the next 4 weeks ahead. I currently just go through all projects and check on a column I created and them pull the data in a report, but my main data is getting too extensive and I'd like to have some functions or automations to do that. I've tried many things but haven't got to the right solution and always end doing manually.
For clarification: What are the license requirements to see the Workload heatmap in a Smartsheet project plan ? I was under the impression that a project manager would NOT need an RM license to see this ?
Active learning methods automatically adapt data collection by selecting the most informative samples in order to accelerate machine learning. Because of this, real-world testing and comparing active learning algorithms requires collecting new datasets (adaptively), rather than simply applying algorithms to benchmark datasets, as is the norm in (passive) machine learning research. To facilitate the development, testing and deployment of active learning for real applications, we have built an open-source software system for large-scale active learning research and experimentation. The system, called NEXT, provides a unique platform for real-world, reproducible active learning research. This paper details the challenges of building the system and demonstrates its capabilities with several experiments. The results show how experimentation can help expose strengths and weaknesses of active learning algorithms, in sometimes unexpected and enlightening ways.

A simple question: i dont have flat connection, so i run p3d offline. I'm wondering if it worth to buy asn. It seems a must have addon, but i have rex 4 (so texture not needed) and i am wondering if asn worths its price just for weather depiction in offline mode.
Can i download historical data for a certain period of time and run historical "continous" (not static but changing realistically based on historical trends) weather? I' ve read manual and some post on this forum, but still i've not understand hoe historical mode works... is it just a different manual weather or it is something like live weather but for the past?
Also i could take an used license for active sky 2012 for a small price, i understood that it is not meant to be used and not supported for p3d, the question is: any chance it would work in offline mode??
Offline mode is just that, offline. You can download something historically, but it is a 'snapshot' of weather at that time. I don't believe you can have it download a span of times and update as you fly. As such, I'm not sure you will see any advantage, as the perks of ASN are that it depicts active weather. You'd only gain the ability to finely tune a snapshot scenario (winds at certain levels, clouds, etc..) but it would be static. Just my 2 cents.
tks Olli, i was not aware of "dinamic weather" working offline and i think it could probably adequately replace what I called "historical continous weather"... does the weather change in realistic way or you are able to see differences with "live weather" behavior, obviously apart from the fact of being able to check the live time accuracy just looking out the window?
You will love it. It adds a very realistic component to FS. I wouldn't fly without it anymore, offline or online. And since you have REX 4, it will make the sky pop, especially when there are multiple levels of clouds.
when i disconnect ASN send me a message saying that the wiz changed to "manual offiline" is it ok? Does the weather change in this way like you said or have i to look for dynamic mode option in settings?

P.S. My recent experience with Google Sheets errors included basic communications errors in my emulator setup timing, and sharing errors. There is also the possibility of Google throttling over use of a free account. It might not be the sheet, it might be you or your connection.
I can download the spreadsheet to the drive and use it. I am the owner of the spreadsheet. But the shared link inside the app has stopped working, and this is the problem. And google says to hope it can work again. But I've seen people with this problem in the Google community a year ago and haven't come back. So I need to update the app with a new spreadsheet link that will have access to the information through the app again.
But I wouldn't want to risk the new link stop working again. So I would like to put several links in the app, so that if the first one stops working (as it happened with this one), look for the information in the next link.
In the other links there would be only copies of the same spreadsheet.

Hi Maurice, just wondering how you are dealing with re-enrollment in this case? From my understanding it isnt possible to have the deals go through this flow multiple time as associated activities cannot be used to re-enroll in workflows. Appreciate the help as this is exactly where I am stuck!
