To me, Grace Jones is the Queen/King of androgyny, she tiptoes the line between masculine and feminine so perfectly and either way she is absolutely striking to look at. Like Ms Jones, Nu is androgynous, it is neither wholly masculine, feminine or unisex, it creates its own rules about gender and takes facets from both sexes.

Nu (this review refers to the Eau de Parfum) was released in 2001 and was created by Jacques Cavallier under the art direction of Tom Ford. I mentioned in my review of Gucci Rush that everything Tom Ford did whilst at Gucci, YSL and Lauder was pretty much epic, and I stand by that. With Nu, he and Jacques Cavallier created something unique and way ahead of its time.

The pepper becomes stronger and spicier in the heart, and as the coolness of the opening fades, the incense comes along to warm things up. What makes Nu genius is the pairing of the warm incense and spices with a sweet, slightly sharp white floral note. I would swear that there is also a dab or two of iris within the heart, there is a soft powderiness with a mere hint of that earthy routines that is the signature of iris.

Nu has excellent longevity and projection, it lasts for over 24 hours on the skin and after each note finally gives up the ghost and leaves the skin you are left with a warm, slightly sour woody/vanilla hue that feels like a second skin.

Thanks for reviewing this beautiful classic. You have reminded me to wear it more often. I have a bottle, but because of the hot weather, I have not worn it for months. There is now a slight chill in the air, and it is time to rediscover Nu!

Hello, I join you in loving NU. I bought it in 2004 when it was still fairly new on the market and fell madly in love with it. The bath products, especially the black soap and the creamy body lotion are spectacular. I did have to learn to be careful in applying the fragrance because it is so strong that several family members objected. I bought several bottles since I had guessed that it might not be around long. Was that ever a good move!

I am working on a large-ish RNA-protein complex with one protein bound with some conformational heterogeneity.
3DVA followed by clustering has been pretty awesome at yielding a discrete conformation.
The cluster structure is binned (256 from 512), so I scale back up to full size particles using NU-refinement (which seemed to work better than homogeneous refinement).
As I suspect the static part of the complex dominates refinement, I tried to follow the NU-refinement with a local refinement, masking around the protein of interest (with no particle subtraction for now).

However, this led to an overall loss of resolution (as I understand things, that would be likely, as the resolution of the NU is at least partially influenced by the static part), and, more importantly, the map looks worse (see attached).

the input for the NU are the particles and volume from the cluster that looks good, then I pull the blobs from the unbinned original refinement (the one that was used for binning).
Is there a more correct way of unbinning?

I have tried using the gaussian prior setting, and I have reduced the SD two 3 and 2 , but I am still losing definition. I am trying to incorporate particle subtraction now, maybe that will help combined with the rotation/shift constraints.

The only other thing that comes to mind is that you may have some compositional heterogeneity (presence/absence of the subunit of interest) in addition to conformational heterogeneity. In this case you might get some benefit from isolating a higher occupancy set of particles by performing masked classification without alignment in relion or similar.

Also what initial lowpass are you using in local refinement? For small volumes, going just a bit worse than the global refinement can be beneficial (e.g. 8 filter for somewhere that previously had an estimated local resolution of 4.5)

At this point, local refinement around my protein of interest is causing the problems I mentioned (as shown in the figure on the right side - all density of my protein of interest just looks worse), and resolution is stuck at 5.8 .

Hi Luca - you may find that a masked classification without alignments does a better job of separating your apo from your holo classes (perhaps as a second step after heterogeneous refinement and an initial NU-refine step)

Resolution is 5.8 , but it is pretty clean (before it supposedly was 5.6, but not really interpretable) . I can trace the whole backbone of the protein-of-interest, and I see blobs for larger residues that help adjusting the crystallographic model I am using.

Now I have finally landed on a concept that works very well for me and my excursions.
