Smart -3SEQ Protocol for single cells

Dear all,

I would like to ask if someone used the Smart-3SEQ protocol for single cells or very low input materials? we are trying to use the protocol to create library from few single cells (50 to 100 cells), but the final concentration of the library that we got was very low (10 picogram in total). Is there any modified version for single cell application or experience in this issue?

How many PCR cycles did you apply?
perhaps try 20?

The laser capture material protocols should qualify as low input. But they are not much different from the original.
