Immune responses during infection, injury, and cancer proceed in the presence of tissue injury and cell death. Consequently, the system must deal with its own dead cells while it determines the appropriate response to the invader. As apoptotic cells are known to induce immune tolerance and necrotic cells can be potent stimulators of immunity, this decision becomes more complex. The key to understanding the immunologic choices made during cell death is to examine the mechanisms of tolerance induction by dying cells and then relate them to the mechanisms of immunity. Ideally, immunogenic cell death should be directed toward tumor cells and infected cells, whereas tolerogenic cell death should be associated with preventing unwanted immune responses to self. In this review, we discuss how the decision is made by focusing on the biochemical process of cell death and how its key components can influence both tolerance and immunity.

Response-adaptive designs allow the randomization probabilities to change during the course of a trial based on cumulated response data so that a greater proportion of patients can be allocated to the better performing treatments. A major concern over the use of response-adaptive designs in practice, particularly from a regulatory viewpoint, is controlling the type I error rate. In particular, we show that the naïve z-test can have an inflated type I error rate even after applying a Bonferroni correction. Simulation studies have often been used to demonstrate error control but do not provide a guarantee. In this article, we present adaptive testing procedures for normally distributed outcomes that ensure strong familywise error control by iteratively applying the conditional invariance principle. Our approach can be used for fully sequential and block randomized trials and for a large class of adaptive randomization rules found in the literature. We show there is a high price to pay in terms of power to guarantee familywise error control for randomization schemes with extreme allocation probabilities. However, for proposed Bayesian adaptive randomization schemes in the literature, our adaptive tests maintain or increase the power of the trial compared to the z-test. We illustrate our method using a three-armed trial in primary hypercholesterolemia.

Q. Traditionally, attacks on data or data infrastructure have been met with similar cyber retaliations. What level of intrusion would give rise to the justification of an armed response?
A. The traditional view has been that attacks with direct consequences in the material world justify an armed response, which is sometimes called a kinetic response. Think of taking down airplanes, causing car crashes, or producing infrastructure failures that lead to immediate death and destruction. Most people believe that a state legitimately can invoke its right to self-defense to respond to such actions with armed force.

The experts who studied these questions on behalf of the North Atlantic Treaty Organization reached that conclusion, but there were dissents then and a few governments have issued statements indicating they might be open to an easier standard. The United Kingdom, for example, has suggested that an attack on its financial system, even without direct physical destruction of people or property, might justify an armed response, if the economic damage is great enough.
Q. Is law regarding armed conflict keeping pace with rapid technology accelerations? Or are technological advances, including artificial intelligence, happening too fast for the law to keep pace?
A. Two norms that seem to me both benign and reachable are: one, no state sponsorship of ransomware or comparable malign cyber-based activity; and two, no armed responses to attacks on big data that do not entail direct physical injury to people or property.

Police is essentially an unarmed service and there is determination both within Police and in the public to keep it that way. But the AOS provides Police with the means of effectively and more safely responding to and resolving situations that pose a high risk.
The basic methods of operating are to cordon, contain and appeal to armed offenders. These tactics are successful in the vast majority of incidents, which are resolved without the use of force. RIVERSIDE (WRGT) - The Mad River School District is teaming up with Riverside Police to add a new layer of security. The board of education has approved a policy allowing school buildings to have an armed response team and locked biometric safes.
According to district leaders, the new safety plan has been in the planning stages for more than a year. The armed response team, which will be made up of volunteers, will go through training before the 2017-2018 school year. Those trained volunteers will then have access to weapons while they're on duty at school. The weapons will be locked inside a biometric safe during the school day. Officers with the Riverside Police Department will determine the location of the safes in strategic areas of the building. The district says only trained members of the response team and law enforcement will know where the safes are and will have access to them.
