Lyme disease, the multisystem illness caused by the tick-borne
spirochaete,
Borrelia burgdorferi, has emerged as a threat to public health
worldwide. It is
a particularly vexing problem in the United States where it is growing
in range
and intensity. In fact, in some hyperendemic regions of New York and
New
England, Lyme disease is now such a threat that it interferes with all
sorts of outdoor activities, and has even led to
depreciation of real estate values. Family dogs in these areas seem to
have been
particularly hard hit by a near epidemic of lameness caused by Lyme
arthritis.
Persons at high risk for infection, such as outdoor workers, campers
and hikers,
suburbanites with lawns to cut, and pregnant women exposed to
potentially
infected Ixodes ticks, are clamouring for some means of protection
beyond simple
behaviour modification and tick avoidance which are known not always
to work.
Hence, the interest in human and veterinary vaccines against Lyme
disease is
growing.

Over 80% of reported cases of Lyme disease in the United States
occur in
coastal regions of northeastern and mid-Atlantic states. The genetic
structure
of the Lyme disease spirochete (Borrelia burgdorferi) and its main
tick vector (Ixodes scapularis) was
studied concurrently and comparatively by sampling natural populations
of I.
scapularis ticks along the East Coast from 1996 to 1998. Borrelia is
genetically
highly diverse at the outer surface protein ospC. Since Borrelia is
highly
clonal, the ospC alleles can be used to define clones. A newly
designed reverse
line blotting (RLB) assay shows that up to 10 Borrelia clones can
infect a
single tick. The clone frequencies in Borrelia populations are the
same across
the Northeast. On the other hand, I. scapularis populations show
strong regional
divergence (among northeastern, mid-Atlantic, and southern states) as
well as
local differentiation. The high genetic diversity within Borrelia
populations
and the disparity in the genetic
structure between Borrelia and its tick vector are likely consequences
of strong
balancing selection on local Borrelia clones. Demographically, both
Borrelia and
I. scapularis populations in the Northeast show the characteristics of
a species that has recently expanded from a population
bottleneck. Major geological and ecological events, such as the last
glacial
maximum (18,000 years ago) and the modern-day expansion of tick
habitats, are
likely causes of the observed "founder effects" for the two organisms
in the
Northeast. We therefore conclude that the genetic structure of B.
burgdorferi
has been intimately shaped by the natural history of its main vector,
the
northern lineage of I. scapularis ticks.

Exposure to Alternaria alternata in US Homes Is Associated With Asthma
Symptoms

Summary
Alternaria alternata is a saprophytic mold typically found in soil and
plants, and is considered to be primarily an outdoor allergen. It has
been
associated with episodes of severe, life-threatening attacks of
asthma, and
sensitivity to Alternaria (ie, as demonstrated with a positive allergy
skin
test) has
been associated with an approximate 200-fold increase in the risk of a
life-threatening asthma attack.[1] Sensitization to Alternaria has
also
been found to be more common in patients with asthma than in those
without
asthma.
Salo and colleagues collected data as part of the National Survey of
Lead and Allergens in Housing study, and they surveyed a nationally
representative sample of over 800 housing units inhabited by over
2400
individuals.
They collected dust samples from bed, sofa, or chair, and from the
bedroom,
living
room and kitchen floors, and analyzed for the presence of Alternaria
using a
polyclonal anti-Alternaria alternata inhibition assay.
They found a positive correlation between the levels of Alternaria in
a
given indoor environment, and the development of asthma symptoms in
those
inhabiting that environment. Of interest, they did not find such
correlation
between this mold and the incidence of allergic rhinitis (hay fever).
They
concluded
that exposure to Alternaria in an indoor environment is associated
with
active asthma symptoms.
Viewpoint
This is an important study for several reasons. First, it builds on
the
increasing evidence that all allergens are not created equal. Namely,
that some (eg, pollens) are more likely to cause symptoms in the upper
airway
(ie, allergic rhinitis), and some (such as Alternaria) are more likely
to
cause symptoms in the lower airway (ie, asthma). Thus, allergy to
Alternaria
is more likely to cause asthma symptoms than symptoms of allergic
rhinitis. The latter has also been suggested by Gergen and Turkeltaub.

Second, the study demonstrates that Alternaria is an important indoor
allergen. Given this allergen's reputation as a potent asthma
trigger,
we now need to be aware that indoor exposure is also important for
patients with asthma.

Perinatal exposure to infectious agents and toxins is linked to
the
pathogenesis of neuropsychiatric disorders, but the mechanisms by
which
environmental triggers interact with developing immune and neural
elements to
create neurodevelopmental disturbances are poorly understood. We
describe a
model for investigating disorders of central nervous system
development based on
neonatal rat infection with Borna disease virus, a neurotropic
noncytolytic RNA
virus. Infection results in abnormal righting reflexes,
hyperactivity,
inhibition of open-field exploration, and stereotypic behaviors.
Architecture is
markedly disrupted in hippocampus and cerebellum, with reduction in
granule and
Purkinje cell numbers. Neurons are lost predominantly by apoptosis, as
supported by increased mRNA
levels for pro-apoptotic products (Fas, caspase-1), decreased mRNA
levels for
the anti-apoptotic bcl-x, and in situ labeling of fragmented DNA.
Although
inflammatory infiltrates are observed transiently in
frontal cortex, glial activation (microgliosis > astrocytosis) is
prominent
throughout the brain and persists for several weeks in concert with
increased
levels of proinflammatory cytokine mRNAs (interleukins 1alpha, 1beta,
and 6 and
tumor necrosis factor alpha) and progressive hippocampal and
cerebellar damage.
The resemblance of these functional and neuropathologic abnormalities
to human
neurodevelopmental disorders suggests the utility of this model for
defining
cellular, biochemical, histologic, and functional outcomes of
interactions of
environmental influences with the developing central nervous system.

NASA USES SATELLITE DATA TO DETECT LYME DISEASE RISK
Scientists at NASA's Ames Research Center,
Mountain View, Calif., and the New York Medical College
(NYMC) in Valhalla, N.Y., are using satellite remote
sensing and computer technology to predict the risk of
Lyme disease transmission.
"We have successfully used satellite imagery to
identify specific types of suburban areas in
Westchester County, New York, where the risk of
exposure to Lyme disease is high," said Sheri Dister,
a research scientist. Dister, with Byron Wood and
Louisa Beck, all employees of Johnson Controls World
Service, Inc., collaborate on the project at Ames.
The NASA team, with Dr. Durland Fish of NYMC and
Westchester County Health Department investigators,
combined Landsat imagery and Geographic Information
System (GIS) technology, which uses layers of maps to
display spatial relationships to map landcover for all
of Westchester County to identify types and placement
of landscape elements associated with Lyme disease
risk.
They found that the higher the proportion of
vegetated residential area next to woods within a
municipality, the higher the transmission risk. It has
been reported that 69 percent of the deer tick bites in
Westchester County are acquired by people near their
homes.
Lyme disease is transmitted to humans by infected
deer ticks. It is now the most commonly reported
vector-borne (transmitted by a variety of insects and
ticks) disease in the United States. When untreated,
it can result in debilitating arthritis, and
neurological and cardiac disorders.

Waxman seeks climate inquiry evidence

The Democratic chairman of a House panel examining the government's
response to
climate change said Tuesday there is evidence that senior Bush
administration
officials sought repeatedly "to mislead the public by injecting doubt
into the
science of global warming."
Rep. Henry Waxman (news, bio, voting record), D-Calif., said he and
the top
Republican on his oversight committee, Rep. Tom Davis of Virginia,
have sought
documents from the administration on climate policy, but repeatedly
been
rebuffed.
"The committee isn't trying to obtain state secrets or documents that
could
affect our immediate national security," said Waxman, opening the
hearing. "We
are simply seeking answers to whether the White House's political
staff is
inappropriately censoring impartial government scientists."
"We know that the White House possesses documents that contain
evidence of an
attempt by senior administration officials to mislead the public by
injecting
doubt into the science of global warming and minimize the potential
danger,"
Waxman said.
Administration officials were not scheduled to testify before the
House
Oversight and Government Reform Committee. In the past the White House
has said
it has only sought to inject balance into reports on climate change.
Present Bush has acknowledged concerns about global warming, but
strongly opposes
mandatory caps of greenhouse gas emissions, arguing that approach
would be too
costly.
Waxman said his committee had not received documents it requested from
the White
House and other agencies, and that a handful of papers received on the
eve of
the hearing "add nothing to our inquiry."
Two private advocacy groups, meanwhile, presented to the panel a
survey of
government climate scientists showing that many of them say they have
been
subjected to political pressure aimed at downplaying the threat of
global
warming.
The groups presented a survey that shows two in five of the 279
climate
scientists who responded to a questionnaire complained that some of
their
scientific papers had been edited in a way that changed their meaning.
Nearly
half of the 279 said in response to another question that at some
point they had
been told to delete reference to "global warming" or "climate change"
from a
report.
The questionnaire was sent by the Union of Concerned Scientists, a
private advocacy group. The report also
was based on "firsthand experiences" described in interviews with the
Government
Accountability Project, which helps government whistleblowers,
lawmakers were
told.
At the same time, Sen. Barbara Boxer (news, bio, voting record), D-
Calif.,
sought to gauge her colleague's sentiment on climate change. She
opened a
meeting where senators were to express their views on global warming
in advance
of a broader set of hearings on the issue.
Among those scheduled to make comments were two presidential hopefuls
- Sens.
John McCain (news, bio, voting record), R-Ariz., and Barack Obama
(news, bio,
voting record), D-Ill. Both lawmakers favor mandatory reductions in
greenhouse
gas emissions, something opposed by President Bush, who argues such
requirements
would threaten economic growth.
The intense interest about climate change comes as some 500 climate
scientists
gather in Paris this week to put the final touches on a United Nations
report on how warming, as a result of a growing concentration of heat-
trapping
gases in the atmosphere, is likely to affect sea levels.
They agree sea levels will rise, but not on how much. Whatever the
report says
when it comes out at week's end, it is likely to influence the climate
debate in
Congress.
At the Waxman hearing, the two advocacy groups said their research -
based on
the questionnaires, interviews and documents obtained through the
Freedom of
Information Act - revealed "evidence of widespread interference in
climate
science in federal agencies."
The groups report described largely anonymous claims by scientists
that their
findings at times at been misrepresented, that they had been pressured
to change
findings and had been restricted on what they were allowed to say
publicly.
The survey involved scientists across the government from NASA and
the
Environmental Protection Agency to the department's of Agriculture,
Energy,
Commerce, Defense and Interior. In all the government employees more
than 2,000 scientists who spend at least some of their time on
climate
issues, the report said.
