Sorry about missing the meeting Monday – I actually forgot what day of the week it was.

 

Anyway, the initial NIST Gaithersburg dataset is uploaded. See the ‘Notes’ doc in the folder for a key to the fcs files.

We had very low densities for the two GFP-producing strains. So, some adjustment might be necessary.

 

Also, we had some issues with intermittent weird scattering signals (tiny bubbles?) that overlapped partially with the e. coli population on the SSC vs. FSC plots. So, some further optimization probably required with our autosampler protocol.

Hi, David:

I've done a first pass with the data that you submitted, and ran into some related issues to the ones that you reported, plus a couple of others:

- Your SSC-H is badly clipping / saturated low
- There are way too few bead points: peaks only at ~100, very ragged; as a result, I think I may be getting the ID wrong on peaks.
- Lots of non-fluorescing events in the sample
- Looks like the 1091 sample essentially failed
