Most of the work I've done up to this point is lab-based DNA assembly.  This is the area I think I can best contribute to.

Two areas I'm interested in are metrics for comparing assembly techniques (i.e. the published protocols and variations for various techniques), and standard DNA toolkits to facilitate evaluating assembly techniques (especially the numerical limits of a technique).

These are important both for general utility, and for specific labs (we all know that handling and equipment varies between labs and personnel).

Toward the second point (standard toolkits), I'm sharing a Google Doc, "A standard toolkit to validate DNA assembly methods", with the group.  It is a proposal to build a broadly applicable standard toolkit of parts which will enable labs to validate a large variety of assembly techniques while generating identical, or nearly identical, final constructs.

It's also attached as an ODT in case the sharing fails.

Feedback and input is GREATLY appreciated!!!!!

Thanks,
