how to speed up barcode QC'ing of new pro strains

Hi,

Idea: How about mating the new pro strains to one or two of the Donors and Recipients in the pilot batch of strains?  Diploids can be easily selected that way and we won't have to waste time making all the pro strains into deletion strains. 

Of course we would never use the strains generated but we can have a relatively quick answer as to which pro strains we want to keep and use to generate barcoded deletion strains.

Thoughts?  If you like it, I can mate our four 384-well plates of pro strains as soon as they grow up.

Hi Atina,

I think it's OK. It can give us an idea on what strains we shouldn't put more effort since the beginning (i.e. those amplifying very poorly (or strongly) after BFG).

However, I'd suggest Marinella and I keep going generating the haploid double mutants the long way, just in case we are not 100 satisfied with the diploids results. We already know which barcodes of Donors and Recipients from the pilot project amplified evenly (Joe's heatmaps) and I'd like to do array-based crosses vs. a few of those.

Joe - for this could you please send us your heatmaps in a table format to know which strains rank close to the Mean/Median?
