I have seen these events listed and have wondered what they are all about.

Simple... Usually a street ride, everyone starts out at one point and is
given a card (playing card). The ride is mapped out and has four check
points, at each check point you are given another playing card. At the
end, the rider(s) with the best poker hand wins!
It can be a lot of fun.

In my part of the country a poker run is sort of like an organized trail
ride. You ride a pre-defined trail loop (arrowed course through the woods)
and draw a card at the completion of each lap. At the end of 5 (or 7)
laps, who ever has the best poker hand is the winner. These are not
contests of speed, although there is usually a time limit to complete the
required laps.
It is a fun, low pressure way to get ready for off-road racing. Also, a
good way to start out the kids.

Or, alternatively, it can be on the dirt. ;) At each of 7 checks, the
rider *selects* a card from a blind deck. At the end, the best 5 cards
are played and the best hands win. The hands are usually pretty un-freakin-
believable. There is a local PR at Rampart they use to raise $$ for trail
maint. I go every time I get a chance, just to support the cause. I had
4 queens, ace high, and only took third. I was just happy to win something.
I usually have two cards that match; like two diamonds or something.

Our club does them off-road. Sometimes you have a map, sometimes written
instructions.
Now and then we do "gunsight" poker runs. You find the next check by
sighting over ribbon tied to bushes to see a landmark several miles away.
(this works in the desert. I don't think that the woods riders would
find it as easy.)
We've also done night poker runs. Nothing like cruising around the
desert on a dark night using a flashlight for a headlight! The checks
were illuminated with traffic flashers, which become visible if you get
within a half-mile or so.

Cool... I had never heard of an off road Poker Run, nor drawing seven
cards (I must get out more). I will take this to some locals and see if I
can't get one going around here somewhere! Just when you thought you knew
everything about the universe... ;-)
The local Honda shop (Lee's Honda) puts one on up here in Redding, CA every spring and it is one
of the most popular rides all year. They actually make it like one loop of an Enduro, about 40-50
miles of trails and 5 checkpoints where you draw one card each time. No time kept so riders of
all abilities show up. You have an Enduro scorecard on which they write down the card you drew.
When you finish the loop - turn in the card and see who wins the prizes!
