If I have an audio file I have brought into an Audacity project, and I have selected various areas where I have generated silence, can I at at later time restore the audio that was silenced in a specific section? Not in the exported file, of course, but in the project.

Cyclin-dependent kinase 9 (CDK9) promotes transcriptional elongation through RNAPII pause release. We now report that CDK9 is also essential for maintaining gene silencing at heterochromatic loci. Through a live cell drug screen with genetic confirmation, we discovered that CDK9 inhibition reactivates epigenetically silenced genes in cancer, leading to restored tumor suppressor gene expression, cell differentiation, and activation of endogenous retrovirus genes. CDK9 inhibition dephosphorylates the SWI/SNF protein BRG1, which contributes to gene reactivation. By optimization through gene expression, we developed a highly selective CDK9 inhibitor (MC180295, IC50 = 5 nM) that has broad anti-cancer activity in vitro and is effective in in vivo cancer models. Additionally, CDK9 inhibition sensitizes to the immune checkpoint inhibitor α-PD-1 in vivo, making it an excellent target for epigenetic therapy of cancer.

In this paper we explore the digital security experiences of marginalised populations in Lebanon such as LGBTQI+ identifying people, refugees and women. We situate our work in the post-conflict Lebanese context, which is shaped by sectarian divides, failing governance and economic collapse. We do so through an ethnographically informed study conducted in Beirut, Lebanon, in July 2022 and through interviews with 13 people with Lebanese digital and human rights expertise. Our research highlights how LGBTQI+ identifying people and refugees are scapegoated for the failings of the Lebanese government, while women who speak out against such failings are silenced. We show how government-supported incitements of violence aimed at transferring blame from the political leadership to these groups lead to amplified digital security risks for already at-risk populations. Positioning our work in broader sociological understandings of security, we discuss how the Lebanese context impacts identity and ontological security. We conclude by proposing to design for and with positive security in post-conflict settings.
A new biography of Shostakovich that views him through the intimate music of his string quartets
Most previous books about Dmitri Shostakovich have focused on either his symphonies and operas, or his relationship to the regime under which he lived, or both, since these large-scale works were the ones that attracted the interest and sometimes the condemnation of the Soviet authorities. Music for Silenced Voices looks at Shostakovich through the back door, as it were, of his fifteen quartets, the works which his widow characterized as a "diary, the story of his soul." The silences and the voices were of many kinds, including the political silencing of adventurous writers, artists, and musicians during the Stalin era; the lost voices of Shostakovich's operas (a form he abandoned just before turning to string quartets); and the death-silenced voices of his close friends, to whom he dedicated many of these chamber works.

My Nest Protect loudly says multiple times a day, last night at 3 am and 6 am, 'SILENCED'. Does anyone know how to silence the Protect? A few days ago my other Nest Protect. Gen2, which is located in a different building and connected to the same Wifi, did activate the smoke alarm and my son silenced that one by pressing the button.
Hi Emerson, thanks for the suggestion but that didn't work. The issue is not that I don't want to silence the alarm from the app but the issue is how to stop the alarm saying so often that it has been silenced. I found another suggestion to thoroughly clean the other Nest Protect gen 2, which appears to have done the trick, because I have not heard my main Nest Protect say that it has been silenced since. Somehow my other Nest Protect 2 must have started to malfunction and constantly trigger the alarm.
