No. They just need to have the same projection.
You can see some typical times for running watershed delineation for
different numbers of DEM pixels and some different machines in the
QSWAT+ manual, section 14.7. So you can estimate a time for your DEM
and decide if you want to resample it. The only recommendation is to
use the best resolution you have if possible.
