Yes. Yes. Yes. and No, not to my knowledge.  :)

We're doing exactly that right now.  We have one stereo backing wav file and one mono click wav file.  Perfectly sync'd.  We have done shows in the past w/ multiple backing tracks, video, midi program changes/cc, and MSC.  Each song was put into a block.  All fire off perfectly everytime.  You can even use a midi pad to fire off each block (each song).

I am looking to find the best platform for backing track playback for a tour coming up.
I need to playback, on cue, a stereo backing track with a seperate click track.
Is Q-Lab that system?
Can Qlab sample-lock multiple tracks together reliably?
Will it work with RME Fireface pre-amps?
or is there a better way to do this?

In your case I think you would want to create a multitrack wave file for each song to make sure everything stays perfectly aligned. In fact, if the stereo file is a band mix, you might allow yourself some additional control by rendering out a multitrack version (kick,snare, stereo drums, bass, guitar L, guitar R, lead vocal, back vocal L, back vocal R, click, etc..) up to 16 tracks that all end up being one file. Easier to manage & would allow for whoever is dealing with the audio some control that a stereo mix doesn't.

To be clear, in the case of a Fireface 800 or 400, "pre-amps" would refer to the mic preamp inputs. Since the questions you have asked all relate to audio playback, you would be using line outputs & not preamp inputs unless you will be mixing live audio with QLab playback.  If you don't have one already, note that the RME Fireface 800 has (8) TRS line outputs & you can also use the headphone output as 9/10 unbalanced outputs. Add (1) or (2) ADAT i/o devices & you can have 8 or 16 more ins & outs. 

If you have an doubts about your concept, you can rent whatever licenses you might need to accomplish your goal by the day to test everything.

QLab is great for this. You'll have to use an app to create multitrack audio files like Sound Files Merger or Audacity.

QLab does not do live audio input. It works flawlessly with the RMEs for playback.

Hi jeremy do you have a link for soundfilesmerger as google doesnt help?

I use a little freebie called de-interleave to split and join files and then audacity when i need to check or edit them

Tried De-Interleave a while back, and for some forgotten reason liked SoundFilesMerger better. But whatever does the job!
