Ah, right, it’s time to break out my annual post :-D
To the best of our knowledge, based on our preliminary testing, QLab 3 works fine on 10.12. Generally speaking, though, we strongly advise waiting until the .2 version of any new OS release if at all possible for any show critical computers. Historically, Apple’s .0 releases tend to be fairly buggy, so it’s important to wait for them to fix the bugs that inevitably pop up when the OS is first widely released.
(Think back just a year ago to the slew of audio driver issues that took a few updates from both Apple and manufacturers to straighten out.)
QLab 2 remains incompatible with 10.11 or 10.12; it is only supported on 10.5.8-10.10.5.
We have had a single anecdotal report of trouble relating to video in 10.12, but it’s thus far unsubstantiated, and may be something completely unrelated to the OS update.
If anybody does run QLab on 10.12.0, and does run into trouble, please email us at sup...@figure53.com with as much detail as possible so that we can investigate.
But, again, if at all possible, give Apple a few updates to smooth out the finish.

The trouble with saying “latest driver” is that it’s always possible for an update to come out without you knowing, or for you to send the email on one day, and for RME to release an update the following day.
According to RME’s support forums, the latest driver is version 2.19_2, and the latest version of TotalMx FX is 1.35. Are those the versions you’re using?
