Recently a descendant of Christoper Poe took part in the Poe DNA project. Out of 37 markers, he differed by three markers from a descendant of Patrick Poe of Bucks Co, PA who married Abigail Day and was a tailor in Plumstead Township in 1721.

It is not possible to say definitively that Chistopher Poe was also a descendant of Patrick Poe, but I can say that out of 37 markers, I differ by FOUR markers from a proven fifth cousin. So I would say there is a strong likelihood that Christoper Poe is a descendant of Patrick Poe. We can only know for certain once more men in this line participate in the Poe DNA project.
A few interesting points:
1. Christoper Day was the father of Abigail Day who married Patrick Poe. The Christoper Poe of Johnson County, IN named a daugher Abigail (I've not seen the names Christopher or Abigail in any other Poe line of that era).
2. Christopher Poe's family has good reason to believe that he was the son of a John Poe who migrated from the Guilford County, NC area in the earily 1800s (the dates differ by account), traveling through Kentucky, and possibly Ohio before settling in Johnson Co, IN.
3. Another family from Guilford County, NC, that moved to Parke County, IN and who are clearly in the line of Simon Poe (1707/8-1793) of Chatham County, NC had close contact with the family of Christoper Poe. Many from both lines can be seen in a series of reunion photos from 1916/17 and were associated with the King family (photos are labeled - King Family Reunion).
Note of caution: the reason there has been so much discussion over the years is confusion over the identity of the John Poe who was in Johnson Co, IN and was alleged to be a Revolutionary War patriot. The DAR records mix several accounts of different men named John Poe. The John Poe who enlisted in Winchester, VA and got land in Stark / Chillicothe County, OH is a different person, who never moved to Indiana. That John Poe was of German descent and was from the Winchester, VA area. I do not know the Revolutionary War history of the John Poe who moved from NC to Johnson Co, IN
