Yolk-sac tumours (YSTs), a germ cell tumour subtype, occur in newborns and infants as well as in young adults of age 14-44 years. In clinics, adult patients with YSTs face a poor prognosis, as these tumours are often therapy-resistant and count for many germ cell tumour related deaths. So far, the molecular and (epi)genetic mechanisms that control development of YST are far from being understood. We deciphered the molecular and (epi)genetic mechanisms regulating YST formation by meta-analysing high-throughput data of gene and microRNA expression, DNA methylation and mutational burden. We validated our findings by qRT-PCR and immunohistochemical analyses of paediatric and adult YSTs. On a molecular level, paediatric and adult YSTs were nearly indistinguishable, but were considerably different from embryonal carcinomas, the stem cell precursor of YSTs. We identified FOXA2 as a putative key driver of YST formation, subsequently inducing AFP, GPC3, APOA1/APOB, ALB and GATA3/4/6 expression. In YSTs, WNT-, BMP- and MAPK signalling-related genes were up-regulated, while pluripotency- and (primordial) germ cell-associated genes were down-regulated. Expression of FOXA2 and related key factors seems to be regulated by DNA methylation, histone methylation / acetylation and microRNAs. Additionally, our results highlight FOXA2 as a promising new biomarker for paediatric and adult YSTs.

The Wenatchee School District is hosting a job fair for bus drivers on Saturday, March 9, from 9 AM to noon at the transportation department building, located at 1201 Millerdale Ave, Wenatchee. The job fair aims to fill open positions for drivers and substitute drivers.


Additional drivers are needed to cover for staff on leave and to ensure that there are enough drivers available for the upcoming busy spring sports and field trip season and into the next school year. Jennifer Sea, the district's transportation director, said the district is looking to fill a training class as soon as possible.


During the job fair, attendees can apply in person, meet drivers and transportation staff, and even test drive a bus. "We wanted to provide people with a chance to test drive a career as a driver. By providing them an opportunity to get behind the wheel of a school bus and drive a little ways in our parking lot, they can get a sense of what it feels like," said Sea.

Physical Demands


REACHING: Reaching is performed continuously; up, down and forward. Work-

ers reach above 72" occasionally to turn on/off oven controls, change prices

on sign, and lift and lower objects to and from shelves. Workers reaching down

to perform such tasks as scooping cornmeal from a plastic barrel, or washing

dishes. Workers reach forward when obtaining toppings, ingredients, cleaning

work surfaces, or answering phones.


HAND TASKS: Eye-hand coordination is essential. Use of hands is continuous

during the day. Frequently activities require use of one or both hands. Shaping

pizza dough requires frequent and forceful use of forearms and wrists. Workers

must monipulo1e a pizza peel when removing pizza from the oven, and when

using the rolling cutter. Frequent and/or force of pinching is required in the as-

sembly of cardboard pizza boxes. Team members must be able to grasp cans,

the phone, the pizza cutter and pizza peel, and pizza boxes -


MACHINES, TOOLS, EQUIPMENT, WORK ALDS: Team members may be

required to utilize pencils/pens, computers, telephones, calculators, TDD equipment, pizza cutter and pizza peel.

Essential Skills

Navigational skills to read a mop, locate addresses within designated delivery

area. Must navigate adverse terrain including multi-story buildings, private

homes, and other delivery sites while carrying product.


CLIMBING: During delivery of product, navigation of five or more flights of stairs may be required.

EXPOSURE TO: Varying and sometimes adverse weather conditions when delivering product, driving and couponing.

SENSING: Far vision and night vision for driving.
