I’m doing some work in the quarter berth of my 10m, removing the old foam-backed vinyl and replacing it with EVA foam. On the cockpit side there’s this molded access hole, about 9”X9”, into the engine bay. 

Couple questions: What’s it for, and what would normally cover it?

I think it's just what you said...port side access to the engine bay.

Havig said that, during my 18 years of ownership...I've only ever used it to access the port side cockpit scupper thru-hull.

Every couple of years, I [blindly] disassemble the thru-hull valve...clean it...grease it...and [blindly] reinstall it...

I don't think anything "covers" that opening...

The PO/original owner cut out a panel almost as big as the cockpit side of the quarter berth, assuming for access to the engine bay, and put a panel of starboard or something similar. Held in place by the cushion. 
I also put a 8” round access port into the roof of the quarter berth to get to the screws at the aft ends of the jib tracks when I was recoring the deck.

I’m wondering how much “air” the engine needs. I would like to enlarge and close mine off as well but there is no fresh air intake unless the blower is running?

I got rid of my quarter birth. All it was juice for is more storage. In the previous owner had junk stuffed underneath it in the back of the boat that was totally inaccessible. So I cut it out and made it cover for the hole that went between the inside of the cabin and that area. It made it much more possible to access the engine the rudder and the rest of the internals for inspection and overall and I was in the yard. 
I don't miss it and it's a lot easier to access below decks in the back. Eventually I may build in one with removable bottom and sides and more space. But it's nice being able to get to all the through deck fasteners and to the through hole for the scupper and I don't miss the extra sleeping space.

I‘ve got a couple of clamshell vents on the cockpit rail and probably plenty of air coming through the locker lids for intake air. No blower with the diesel, though.

I know a lot of folks feel that way. I’ve always liked a quarter berth either on passage or in a rolly anchorage. It’s a nice, snug cocoon if you’re not claustrophobic and can contort your way in!

The opening is present on my #26 10M as well.  I thought it was so someone could reach the port scupper through hull handle.  
The PO of my boat mounted a car radio next to the hole.  Something I need to remove, as I never use it.

But do you have a gas dingy outboard, propane stove, or portable generator? Any of these and you STILL need to have a blower for safety.
