How should I set the output frame?

As I know, before, on a traditional animation paper, there is a box called 'output frame'. Animators usually drew some part out of the output frame.

However, just now, when I opened OpenToonz, I did not see any output frame on the canvas, and I don't know how to set it.

It seems like you're talking about having a canvas that you can draw on that is larger than is shown on the cameras. If so, then that's easy to solve. When you create your levels (by clicking the New Level buttons on the timeline toolbar or from the Level menu). just type in a size that is larger than your camera size.

Just now, I found the 'safe area' function in OpenToonz. What is the purpose of safe area? Will it affect the final output?

No, the safe area won't affect the output. It's a grid to show you the safe area where you should draw within for traditional TV or cinema viewing, so that the important parts of your drawing aren't cropped out. For release on the internet, this really isn't important.

I'm not sure why you'd want the camera size to be the same as the safe area. It doesn't really serve any purpose apart from making your image smaller. But if you want to change the camera size, you can do so when you create your project or, once created, change it in the menu, xsheet > Camera settings.
