Hi,
I am Naty. First of all, thank you for this great tool for analyzing sperm trajectories and more.I was using the OpenCASA V2 plugin in Fiji very successfully.

However, right in the middle of my batch of videos, it suddenly stopped identifying any sperms. Here are the details of the issue. The Problem: Fiji opens the .avi videos perfectly and completes the process, but the final output videos in the results folder show no trajectory lines at all, and no Excel sheets are generated.
What I have tried so far: I already changed the memory and thread settings, uninstalled and reinstalled the plugin and fiji and imageJ, and checked my configurations, but nothing seems to fix it.
The videos are identical. To help diagnose the issue, I have uploaded the two videos here: Successfully tracked video
I would deeply appreciate any guidance or help you can provide to find out what is causing this.

Best regards,
Naty

Hi Naty, welcome!

First of all, have you been able to analyze videos before?

By your description, it looks like you need to follow these instructions
(you can find them in the introduction to the group):

OpenCASA v. 2 does not work if directly installed on recent versions of
ImageJ/Fiji. This is the "NA error." If you try analyzing, you only get
NA in the results window.
You need to go to https://wsr.imagej.net/distros/, go to the folder for
your system, and download an older version (1.53k from 2021 should
work). Just unzip 1.53k, run it, install OpenCASA (do not forget
javaml-0.1.7.jar), and run it. Close it and go back to your modern
version (maybe it is a good idea to reinstall the plugin).
You might want to check if Java is up to date on your computer. If it's
not working, try switching between zips, including Java or "nojre" (no
Java).

I have asked for permission for accessing the videos, it seems that the
link is for restricted access.

Best wishes,

Felipe

Hi,
OK, after cheking the videos I understand that OpenCASA yields output
for some of them. Then, it stops processing, am I right?
It happens sometimes. Usually, OpenCASA finds something unexpected in
the folders (folders or files from previous analyses, non-video files,
hidden files, etc.). In this case, even if annoying, the best course of
action is to identify the culprit folder, move it out of the parent
folder and test again. Using the single folder or single file approach
might help identifying "corrupt" folders or videos making OpenCASA stuck.
I suggest that you try this solution first. Sometimes the problem is not
apparent, but a similar situation has happened many times in our lab,
and the solution was cleaning the input folders from previous analysis
files or identifying corrupt videos.
Best wishes,
Felipe
