Tracking human sperm

Hi, My name is Pilar

I'm using openCASA to tack human sperm cells, acquired with a 10x objective, at 100 fps.

I cannot manage to set the parameters to get a correct tracking, and I wonder if anyone can help me. 

The openCASA only detects a few sperm cells and not the entire track, even sometimes it detect one track like it were two diferent cells. 

On the other hand,  I'm not so sure if I making the right video capture. Has anyone a suggestion on how to adquire the video? 

I leave attached my settings parameters and the results of openCASA for one video.

Dear Pilar,

Welcome to the OpenCASA list. Using your setting parameters (even
considering that you might change the frame rate to 100 fps), the
results I obtained ares quite aceptable (please check the image
attached). Probably you may have problems with the ImageJ version or the
windows version (see previous emails in this list).

Hi Jesus! Thank you so much for your answer. 

Like you said, I'm starting to think that I have a problem with the version of FIJI or the OpenCASA plugin cause I tried with a video and parameters I knew that works and no sperm cells were detected. 

I'm worried about this cause I need to install the programs in a new computer for work and I don't know what i'm gonna do

Hi.
I was testing in different computers, and maybe we have a problem with
ImageJ versions.
I have tested with v. 1.53a in MacOS and 1.53e in Windows and it works
(by the way, you have 100 fps in your settings, I believe, should it be
60?).
Maybe this is the same problem Salvador reported. This is quite strange,
since I tested in MacOS with 1.53k (not sure, it was in another
computer!) and your video worked in a first attempt... then I was unable
to replicate it! (even after reinstalling the plugin, updating ImageJ,
resetting IJ options with Edit/Options/Reset). In fact, sometimes it
even failed to produce the output video, sometimes producing it but with
no trajectories (as expected).
My advice is that you tested with 1.53e (if I have some time, I'll tried
with some other versions, just in case). If that's the problem, maybe
this is a minor glitch with the code being incompatible with some of the
upgrades (however, this is beyond my knowledge, no experience with Java).

Hi everyone, I managed to solve my problem! Like you said before it was a problem with the version of imageJ. I delete my version (by the way I don't know what version it was) and now I'm using the 1.53 and it works!!

Now I have to install other plugins and hope it doesn't update the entire app
