Hello people, my name is Alejandro, I have a PhD in Biological Sciences, and I work for an Animal Biotechnology laboratory at the Universidad Nacional de Río Cuarto (Río Cuarto, Córdoba, Argentina). Our group is currently working with biotechnological and bioengineering tools for different animal species, aiming at productive purposes and animal improvement.

For some time now, we have been developing polymeric surfaces to select sperm populations from different species (bovine, equine, and porcine). We have been trying to optimize different software options for sperm quality analysis and came across this software.

I managed to upload short videos (about 1 or 2 seconds) to the software, but I keep getting "NaN" as a result for all the parameters.

I need to determine whether the quality of the videos I used is sufficient. We have a camera attached to the microscope, but it is old, so the quality is not very good. Instead, we have been recording the videos with a cellphone. Additionally, the phase contrast of the microscope is poor.

If someone could help me or give me advice on how to make it work, I would really appreciate it.

Hi Alejandro.
First of all, you have find a well-known glitch in OpenCASA. You need
to go to ImageJ website and download an older version (maybe 1.53g?).
You need to go to https://wsr.imagej.net/distros/, choose your
operative system and download a 2020 version. You run OpenCASA on it
and then you can close it, remove it if you like and go back to the
updated ImageJ or Fiji, then OpenCASA works.
However, your need to improve the video, indeed. The phase contrast is
not good (I have found the same problem with many colleagues, many
brands claim to sell negative phase contrast, but either they are
terrible quality or not real Ph-). The grid is also interfering, you
can just flip the coverslip piece to get it out of focus.
Anyway, I feel that the magnification is too low (the image is much
larger in our microscope; not a big issue, anyway, since you can adapt
OpenCASA to different circunstances).
In any case, we would need some information for replicating your
analysis: microscope model, magnification, fps, scale, etc.
