Settings for consentration module with canine semen.

Hello dear friends, i'm trying to get OpenCASA working for diagnostic use at our clinic and so far i've worked out what seem to be good settings for the motility module but the concentration odule is giving me a lot of trouble. Currently it's giving me results that are 8-10 times higher than the actual amount of spermatazoa and i don't know if it's a setting issue or a problem with my method or equipment.

My hardware consists of:

Windows 10 Pro machine

a 1920x1200@165pfs (adjustable) monochrome camera

Nexcope 610 with a dark field insert.

a Improved Neubauer chamber

All images were taken at 10 times magnification,  and a dilution of 1:100

i've attacheed images taken of thawed semen with a previously recorded concentration of 277 M/ml

Dear Artur,

The images you sent have not enough contrast for a proper analysis. I
recomend you use negative phase contrast microscopy.
