I hope you all are doing well. My name is Dhiraj Puri, and I am a research scholar from BITS Pilani, Goa Campus, India. I am currently working on human sperm cells and need to use the Open CASA plugin in ImageJ for tracking.
I have tried different versions of ImageJ (1.54, 1.53, and 1.49), but I am unable to track the sperm cells successfully. I would appreciate your help in determining whether the issue lies with the video or the plugin.

I have not proccesed the video. It was used directly.
The key points are:
the settings (mainly the microns/pixel and number of frames rate of your camera). In this example I use 0.28 microns/pixel and 50 frames/sec.
the quality of the video,

Are you processing the video before using it in OpenCAsA (such as adjusting thresholds or other settings)?
