Issue related to ImageJ and Video for Open CASA

Hello,

I hope you all are doing well. My name is Dhiraj Puri, and I am a research scholar from BITS Pilani, Goa Campus, India. I am currently working on human sperm cells and need to use the Open CASA plugin in ImageJ for tracking.

I have tried different versions of ImageJ (1.54, 1.53, and 1.49), but I am unable to track the sperm cells successfully. I would appreciate your help in determining whether the issue lies with the video or the plugin.

Dear Dhiraj Puri,

I have evaluated the video you shared, and I can detect the sperm motility in OpenCasa (although I do not have the exact scale of your microscope system to calaculate the microns per pixel, I used 0.2).

Based on my experience with my MSc students, I think you have a problem with the ImageJ version.

Dear Dhiraj Puri,

I have not proccesed the video. It was used directly.

The key points are:

the settings (mainly the microns/pixel and number of frames rate of your camera). In this example I use 0.28 microns/pixel and 50 frames/sec.

the quality of the video,

Thank you all! I have finally been able to track sperm cells. I tested each version available in the distros folder, and version 1.51k worked for me. Additionally, I found that adjusting the contrast of the video—making it slightly darker—improves performance.
