FARSC - Feed Antimicrobial Resistance Selection Concentration
"Concentration (FARSC) study developed specifically for the assessment was applied. However, due to the lack of data on the parameters required to calculate the FARSC for amprolium, it was not possible to conclude the assessment. To address growth promotion,
data from scientific publications obtained from an extensive literature review were used. Levels of amprolium in feed that showed to have an effect on growth promotion/increased yield were reported.
The lack of antibacterial activity at clinically relevant concentrations for amprolium suggests that further studies relating to bacterial resistance are not a priority."

"Conclusions
It is the overall opinion of this commission that the benefits of antibacterial additives fed to Animals do not outweigh the risks.
With regard to medicinal substances, they are specifically used to prevent disease and therefore they should be treated as pharmaceutical specialties

Some chicken grain contain amprolium (not an antibiotic)
Technologies and Risks
amprolium is structurally similar to vitamin B1. When it is administered orally to chickens, it has a minimal bioavailability, but the absorbed fraction is widely distributed into the tissues and rapidly eliminated in the urine and feces. Amprolium administered
to laying hens is deposited primarily in the egg yolk, and residues can be detected in eggs for ~ dozen days or more after cessation of treatment, depending on the dose and assay sensitivity.

blockage of the Thiamine metabolic pathway is perceived as severe deficiency of that Essential B1 Vitamine, or starvation causing Wernickes etc, apparently fatigue, hypoglycemia, &c
Impression is that Patients with the CFS (chronic fatigue syndrome), post covid should avoid amprolium from any source
As mentioned previously, there are some antimetabolites which block the Thiamine pathway, but don't reduce the serum level of that Essential B1 Vitamine
That implies it would be difficult to measure the Thiamine levels or if that pathway is open
Unusual that request to Lab to measure presence of any Thiamine antimetabolites wasn't answered

Thiamine Analogues
Amprolium (Amprol®, Amprolium®, Ampromed®, Amprosid®, Amprosol®, Amprovine®) is a quaternized derivative of pyrimidine that interferes with thiamine metabolism. It is a thiamine antagonist, blocking the thiamine receptors, thus preventing carbohydrate synthesis.
amprolium is defined as a thiamine analog inhibitor that is used as a coccidiostat for chicken feed, which could induce experimental disease

Agricultural and Biological Sciences
A maximum residue limit is defined as the maximum concentration of a compound residue that is allowed in or on a food commodity at a specific stage in the harvesting, storage, transport, marketing, or processing of the food, up to the final point of consumption.

7.3 Toxicological aspects
Coccidiostats are generally considered to be free from side effects and
toxicity as long as they are used in their target species at the correct dosage.
However, chemotherapeutic coccidiostats have a narrow range of safety and
there are many reports in the literature of accidental intoxications with
various anticoccidial drugs. However, most such events can be avoided if
proper manufacturing practices (GMP) are maintained.
Intoxications with coccidiostats have been reported with
poor mixing of the grain resulting in uneven cocccidiostat concentration,
failure to properly dilute the coccidiostat concentrate, and in cases of
incorrect identification of the product (Szancer, 1989; Novilla,
1992). The feed can become cross-contaminated at the production mill and
Animal meal has on occasion been shown to contain traces of coccidiostat.
Toxicity has also been noted when giving poultry meal
containing coccidiostat to animal species for which the feed was not intended
Diagnosis of coccidiostat intoxication is sometimes difficult due to
reversibility of clinical signs and/or the variability of pathological lesions
found with toxicity of coccidiostats of different chemical groups. Feed
analysis is often not possible to perform because the feed has already been
consumed
7.3.3 Residues
To avoid residues of coccidiostats in meat, withdrawal periods before
slaughter of the animals are assigned (table 7.I.). Eggs are, however, a
special problem. Maximum age levels and restrictions on usage in layer feed
may not always be enough to ensure that yolks are free of coccidiostats.

"Comparison of the effects of some thiamine analogues upon thiamine transport through the BBB to the CNS (Central Nervous System)
A series of chemical analogues of thiamine were given to see how each affected the flux of thiamine into the brain and the findings are compared with those for a fifth analogue studied in previous work. Pyrithiamine, thiamine disulphide and acetylthiamine,
like amprolium, inhibited thiamine transport across the BBB
analysis shows that they compete mainly for the saturable component of thiamine flux across the BBB, with only a slight inhibition of the non-saturable component, most clearly seen with pyrithiamine.
oxythiamine, despite its similarity to thiamine did not have any significant effect upon the flux of the vitamin into the brain.
These findings explain the efficacy of pyrithiamine administration, especially in conjunction with a thiamine-deficient diet, in rapidly producing central neurological signs of deficiency.
(PREVIOUS STUDIES FROM THE 1930'S STATE THAT OXYTHIAMINE DOESN'T ENTER THE CNS (CENTRAL NERVOUS SYSTEM) CAUSING ONLY PERIPHERAL DISEASE, BUT PYRITHIAMINE DOES, CAUSING WERNICKES-
However, if the BBB is damaged by real Thiamine deficiency, previous administration of neopyrithiamine, etc, then oxythiamine would enter the CNS through the BBB and cause more serious disease-
oxymoronic scams are counterproductive)
