Hi,

Some deviations are expected because your protein construct is different and the environment is not identical to the reference. First, you may need to delete peaks that do not exist in your protein; then, do your best to manually adjust the locations of well-isolated peaks using the mouse and center them using 'pc.' Further experiments, such as TOCSY-NHSQC, can be helpful to resolve overlapping regions.

단백질 구성(construct)이 다르고 실험 환경이 참조 데이터(reference)와 완전히 동일하지 않기 때문에, 어느 정도의 편차는 예상되는 부분입니다. 먼저 분석 중인 단백질에 존재하지 않는 피크들을 제거해야 할 수도 있습니다. 그 후 마우스를 사용하여 잘 분리된 피크들의 위치를 최대한 조정하고, 'pc' 명령어를 사용하여 중심을 맞춰주세요. 겹치는 영역을 해결하는 데에는 TOCSY-NHSQC와 같은 추가 실험이 도움이 될 수 있습니다.

안녕하세요, 박사님.

저는 NMR 을 배우고 싶어서 독학하고 있는 단백질 구조생물학 전공 대학원생 유현정이라고 합니다.

단백질과 화합물 결합을 2D NMR로 보고 싶어서 15N 라벨링을 해서 단백질을 발현 정제하고 900 MHz 에서 HSQC 데이터를 얻었습니다.

박사님의 유튜브를 참고해서 BMRB 코드를 넣고  assignment 하는 것까지 진행해보았는데, reference와 같은 정제 버퍼를 사용했는데 peak assignment가 조금씩 틀어지는 것을 확인하였습니다.

제 타겟 단백질이 reference 단백질보다 c-terminal에 아미노산 잔기가 44개가 deletion 되어있는데 그것 때문일까요? 어떤 픽들은 전체적으로 조금 이동한 것 처럼 보이는 반면, 어떤 픽들은 아예 보이지 않아 assignment 가 어렵습니다. 또한 여러 픽이 겹쳐 있는 구간에서는 어떻게 assignment 를 진행해야할지 막막하기도 합니다.

기본적인 질문일 수 있겠지만, 도움 받을 곳 없이 혼자 공부하고 있는 입장에서 너무나 막막하여 문의를 드리게 되었습니다.
