I would like to know what is the best way to reject mail from one
particular user that is sending mail to my site.
us...@domain1.com sending mail to my site domain2.com.
My smail configuration uses the smart host logic for mail routing to our
exchange server. I changed the address rewriting rule new_user so that
is rejects a certain user. It works but is there a better way?? If I
have more than one user I want to reject mail from then it would get
messy.

smail doesn't offer any good way to reject email *from* one particular
user. if it's email *to* one particular user, that is easy to dispose
of in an automated fashion.
smail does have provisions for refusing email from particular hosts
or particular domains.
typically people recommend the use of procmail to handle this sort of
issue. the typical approach is to put a procmail invocation in the
.forward file of the target usere in question, but there's some gotcha
about doing that correctly. look at the archives of this mailing list
for more info on that.

you may be more knowledgable about this than i am, since i didn't think
there was any way that the rewriting rules could affect email *from* a
particular user.
