System wide, or just for Thunderbird?

For Thunderbird it is Menu Button > Preferences > General > When new
messages arrive, and select a sound file of your choice for "Use the
following sound file".

I have not tried to change it system wide, but it looks like it can be
changed just for email notification in System Settings > Application and
System Notifications > Applications tab and use the Event source drop
down to find "New email notify". That's what I see using Linux Mint/KDE.

I'd go with changing it in Thunderbird.

If you mean a Calendar Event that is a different support group at
mozilla.support.calendar.

However, Menu button > Preferences > Calendar > Reminders
