Try installing somewhere where you have write (and file-creation) permissions:
either log in as Administrator before you install Thunderbird for everyone (in
its default location) or else (if, for instance, you aren't privy to the
sysadmin password) install it privately (by changing the install directory
during Thunderbird Setup) somewhere under your home directory. Also, make sure
that you haven't got some mozMapi32.dll with readonly attribute leftover
wherever it is that you're trying to install.

I'm on Linux, where Thunderbird is distributed as .tar.gz archives with no
installer. Before I unpack each day's nightly, I just remove the installation
directory of the day before (and all its contents).
On Windows, you should be able to download the self-installer anywhere
convenient and run it from there (as Administrator); it should detect the
previous installation and remove it (keeping your settings and extensions,
which are elsewhere).

did you manage to fix Thunderbird...I have the same error and I am
unable to
get Thunderbird up and running again!

I did uninstall all of my Thunderbird but before this, I discovered
that there is a Mozilla Program backup.
I backed everything up after about 30 minutes and uninstalled it. Now
everything is working after downloading Thunderbird 2
For those of you on Google who're searching and see this, check if
you've installed any Logitech camera software (QuickCam etc). It uses
mozMapi, and won't let Thunderbird delete it to upgrade. All you need
to do is exit the program that sits in the tray and then install
Thunderbird.
Took me a while to figure it out, hope this helps someone else with
the same error.
