All the settings for the email configuration is done in the Administration>Parameters>Email section for bugzilla, but the most common problem is the system settings. Start by checking the bugzilla perl mods are working correctly by setting the 'mail_delivery_method' to 'Test' to make sure the emails look good in the data/mailer.testfile.

Check your path and make sure any directory listed is not all read/writeable by everyone. Especially if the default path has been modified for the user running bugzilla.
Check the postfix logs on the bugzilla server, they will usually indicate the problem, such as the SMTP server doesn't accept emails from your bugzilla server address.
