This looks like a problem on the level of your web server or network.
Did you change anything else besides Bugzilla, e.g. upgraded the web
server or OS or even moved your Bugzilla to another system or
installation directory or such?
I suggest you activate smtp debug in Bugzilla mail settings, try again
and have look in the log of your web server for some more details,
especially in which status of the SMTP connection the error occurs.
I'm not change the web server/OS. I just upgraded the bugzilla that steps are following bugzilla document.
Below am i using steps:
1. download the stable version of bugzilla tarball file.
2. untar it then rename to bugzilla
3. bakup previous bugzilla to bugzilla.bk
4. Into the bugzilla folder and copy the localconfig and data/ to new bugzilla folder.
5. Run the ./checksetup.pl (there have some suffer it need to update the some modules.)

I suggest you check the mail communication manually by using telnet on
the server hosting your Bugzilla to see if any needed libs are
available and such. Sometimes openssl-dev-packages and such are
missing because of insufficiently modeled dependencies.
