1) What was the reason for the addition of CHIR99021 in Phase 1 of the Differentiation Protocol? Just more efficient mesoderm induction and potentially higher likelihood of lymphoid differentiation?

2) Did you already try adding IL-7 or DLL-1 at some stage in Phase 3 to induce lymphoid differentiation?

Yes more efficient mesoderm differentiation 
We have a bunch of lymphoid data... watch this space ;)

Hi Abs,

another question regarding media composition throughout the differentiation protocol: Did you try switching from APEL2 to another (preferably cheaper) medium like StemPro-34 or E6 in the first 14 days of the differentiation protocol?

Hi Karo! 

We haven't tried this, it's been on my to do list for ages but have never found the time. 

I think there are a lot of options for early meso/haem differentiations I'm sure there are some that might be worth trying!

PS You can also get the original APEL recipe from the Ng et al. Nature Protocols paper which may wind up cheaper !
