Beating organoid

Hi everyone! I have a question for the group.

At 33 days of culture, we observed a small area beating organoids.

Has anyone observed beating bone marrow organoids around day 30 of culture? We found that in just two days of culture, the number of beating organoids increases, and we are wondering if others have made similar observations, either at the same time or at different times.

Hi Lea, 

So we haven't seen this, but considering that cardiomyocytes are one of the easier mesodermal lineages you can get out of iPSC diffs it's not a huge surprise! It implies that some of the factors that specify towards our lineages of choice are not working well (e.g. low oxygen, hSCF, FLT3L, longer term BMP4). It might be worth checking your incubator and your cytokine stocks, particularly for factors that are specific to the comBO/BMO protocols (e.g. CHIR is shared across a lot of diff protocols).
