Blocking buffer in IF whole-mount staining

Hi TooT, 

In whole mount IF staining, the UEA primary antibody used is biotinylated (B0-1065-2 Vector Lab). 

Is there a need to use endogenous biotin blocking kit prior to blocking or is the blocking buffer you use containing goat serum, BSA, PBS, Triton X-100, Tween-20 and sodium deoxycholate sufficient to block background ?

Morning Ka Ka, 

The blocking buffer we use (that you've detailed above) is enough for UEA staining - it's not an antibody (it's a lectin), so tends to give a very clean signal.
