Primary cell engraftment using adjusted hydrogel method

Hi all,

did anyone compare the original hydrogel method (12-well plate from D15-D12 of diff protocol) to the new microgranular gel method (directly embedding aggregates on D5 in a 96 well plate) in terms of facilitation of primary cell engraftment?
I am witnessing slightly lower engraftment percentages/numbers after the same number of days of co-culture with the microgranular gel.

Would be very thankful if anyone could comment on their experiences:)

Morning Karo, 

So with the comBO protocol we tend to do the engraftments later (d21 onwards), and get pretty good engraftment rates. At least what we've found is we can titrate the number of donor cells down and get consistent engraftment and expansion (using 1k cells per well at the moment). 

Is there data you can share so I can get a better sense of what you're seeing? Feel free to email if you'd prefer it wasn't on here!
