Lens for V4, 0.5 pitch

Hi everyone,

I’m setting up calcium imaging with the UCLA Miniscope V4 in GCaMP6 transgenic mice and wanted to ask for advice on GRIN lens options.

My goal is to image layer 5 neurons in visual cortex, ideally using a prism-coupled GRIN relay lens. From what I understand, the V4 requires ~0.5 pitch relay lenses, and most standard catalog lenses (e.g., Edmund) are around 0.25 pitch, so they may not be suitable.

I’m currently looking for:

  • ~1.0 mm diameter GRIN relay lens

  • Pitch ~0.5 (compatible with V4)

  • Compatible with a 1 mm prism

  • Suitable for chronic cortical imaging (mouse V1, layer 5)

  • Good transmission for GCaMP6

I would really appreciate any suggestions on possible lens options, specific part numbers, or vendors that have worked well for similar setups. Also, if anyone has experience with prism + relay GRIN combinations in cortex, I’d be very interested to hear about practical considerations.

Hi Somoday,
I would suggest sourcing your GRIN + Prism implant from Inscopix. All their GRIN lenses and GRIN + Prism lenses are fully compatible with the V4 Miniscope.  Building this out yourself would require a specific pitch GRIN lens to push the working distance out past the glass of the prism. This can be done but it is quite difficult to get custom GRIN lenses made nowadays.
