You have ISA on the same box? Is this an SBS box or plain Windows?
You will want to troubleshoot what is being blocked by the ISA.
Ideally they shouldn't be on the same box unless you are using SBS and
in that case there is a specific configuration that the setup does.

Please don't multi post questions, especially with different subjects.
It confuses the heck out of people trying to help. If you are unsure
as to where to post a question use a cross post to send one message to
various candidate groups.

Hi, this is plain windows. I've made a rule in ISA to allow local
connection. What rule is needed for exchange in ISA ? I'm just testing
exchange on the server.

Good question and one I can't answer since I would never allow a
server to be in the configuration you have it in.
Good plan would be to go to the ISA groups and tell them the hardware
config (in terms of what all your NICs are doing/facing) and ask them
how you make sure MAPI (RPC) communications are allowed from the
inside.
