Rebuild the server again and do a backup. Then restore that backup
into the Recovery Storage Group (read about that at
www.microsoft.com/exchange/library) to test your backup.

That will restore the backup into a separate place on your exchange
server without dismounting or doing anything to the store (i'm
assuming the whole thing is in the lab right now?)

Using ntbackup should be all you need, but see also the DR guide (link
as above) and see if you're doing anything differently from that
described in the documentaion.
