Hi,
I have tried to use fluorescence images of FM 4-64 stained cells instead of phase contrast, but so far without luck. It is probably possible to convert the image so that it looks more like a phase contrast image somehow. Anyone here that has any experience with that? It would be very helpful with some advice.

Try to set the invertimage parameter to 1 and that should do it.
